SALSA MLPA KIT P277 Human telomere-10

This P277-A1 Human Telomere-10 probemix can be used to confirm and further characterise abnormalities detected by SALSA MLPA kit P036 Human telomere-3 and/or SALSA MLPA kits P069 / P070 Human telomere. This SALSA MLPA P277 probemix contains: 13 probes in the terminal 6.8 Mb of chromosome arm 5q. 10 probes in the terminal 4.5 Mb of chromosome arm 6q. 10 probes in the terminal 6.8 Mb of chromosome arm 7q. 10 probes in the terminal 4.6 Mb of chromosome arm 8q. SALSA MLPA kits P036 and P069/P070 each contain one MLPA probe for each subtelomeric region. Most of these probes detect a well-characterised gene close to the telomere. The probes in P036 and P069/P070 are different, allowing results obtained with one probemix to be confirmed by the other. The majority of probes in P069 are identical to the P070 probes. However, in contrast to P070, no probes for the acrocentric chromosome arms 13p, 14p, 15p, 21p and 22p are present in P069. We recommend to use SALSA MLPA kit P036 Human telomere-3 for primary screening of the subtelomeres. Results obtained with P036 can be confirmed and further characterised with a follow up kit for specific telomere analysis, such as this P277 Telomere-10 kit. Heterozygote deletions of probe recognition sequences should give a 35-50% reduced relative peak area of the amplification product of that probe. However, mutations and/or polymorphisms very close to the probe ligation site may also result in a reduced relative peak area. Therefore, apparent deletions detected by a single probe always require confirmation by other methods.
